چکیده

Atherogenesis comprises three stages, including intimal thickening, plaque formation and destabilization and rupture of the plaque. Intimal thickening develops depending upon aging and is characterized by increased collagenous matrix. Plaque formation is characterized by an accumulation of intraand extracelluar lipids in intima, resulting in the advanced lesion with atheroma. Finally, advanced lesion is destablilized by plaque inflammation, and subsequently ruptures, leading to clinical manifestations. AGEs include various chemical structures, such as(carboxymethyl) lysine,(carboxymethyl) arginine and pyrraline. Different AGEs are derived from different pathways, which are oxidation-dependent or oxidation-independent. In this paper, the role of various AGEs in different stages of atherogenesisi is described.

Prevalence of Atherosclerotic Plaques in Autopsy Cases with Noncardiac Death

Background and Objective: Ischemic heart disease (IHD) following atherosclerosis is the most common cause of cardiac deaths world wide. We aimed to investigate the pathologic features of atherosclerosis in non cardiac death cases to have an estimate of atherosclerosis prevalence in Mashad (North east of Iran). Patients and Methods : This descriptive (cross sectional) study was done, during ...

Advanced glycation end products and vascular inflammation: implications for accelerated atherosclerosis in diabetes.

The formation of advanced glycation end products (AGEs) is an important biochemical abnormality that accompanies diabetes mellitus and, likely, inflammation in general. Here we summarize and discuss recent studies indicating that the effects of AGEs on vessel wall homeostasis may account for the rapidly progressive atherosclerosis associated with diabetes mellitus. Driven by hyperglycemia and o...

RAGE biology, atherosclerosis and diabetes.

Diabetes is characterized by accelerated atherosclerosis with widely distributed vascular lesions. An important mechanism by which hyperglycaemia contributes to vascular injury is through the extensive intracellular and extracellular formation of AGEs (advanced glycation end products). AGEs represent a heterogeneous group of proteins, lipids and nucleic acids, irreversibly cross-linked with red...

Forum: Role of Oxidation in Atherosclerosis GLYCOXIDATION AND LIPOXIDATION IN ATHEROGENESIS

Atherosclerosis may be viewed as an age-related disease initiated by nonenzymatic, chemical reactions in a biological system. The peroxidation of lipids in lipoproteins in the vascular wall leads to local production of reactive carbonyl species that mediate recruitment of macrophages, cellular activation and proliferation, and chemical modification of vascular proteins by advanced lipoxidation ...

A study of iron status in patients with coronary atherosclerosis

Background: It has been demonstrated that free oxygen radicals have a close interaction with lipid peroxidation, causing a modification of low-density lipoproteins (LDL) and facilitating LDL deposition, with the consequent formation of atherosclerosis plaques. Free radical production is catalyzed and accelerated in the presence of iron. This study was conducted to evaluate the possible associ...

Metformin Inhibits Advanced Glycation End Products-Induced Inflammatory Response in Murine Macrophages Partly through AMPK Activation and RAGE/NFκB Pathway Suppression

Advanced glycation end products (AGEs) are major inflammatory mediators in diabetes, affecting atherosclerosis progression via macrophages. Metformin slows diabetic atherosclerosis progression through mechanisms that remain to be fully elucidated. The present study of murine bone marrow derived macrophages showed that (1) AGEs enhanced proinflammatory cytokines (interleukin-1β (IL-1β), IL-6, an...
